Matrixyl: Dosing Protocols

Important Note#

Matrixyl (palmitoyl pentapeptide-4) is a cosmetic active ingredient applied topically to the skin. It is not an injectable peptide and does not require reconstitution or injection. All application is via topical skincare products.

Robinson et al. (2005) -- Pivotal Clinical Study#

The primary clinical evidence for Matrixyl's efficacy comes from a 12-week study using:

• Concentration: 3 ppm (0.0003%) Pal-KTTKS in a moisturizer base
• Application: Twice daily to one half of the face (split-face design)
• Population: 93 Caucasian women, aged 35-55, with facial photoaging
• Results: Significant improvement in wrinkles and fine lines vs the moisturizer-only control side

Short-Term High-Frequency Studies#

Additional studies using twice-daily application for 28 days have demonstrated:

• 18% reduction in wrinkle fold depth
• 37% reduction in fold thickness
• 21% improvement in skin rigidity

Commercial Product Concentrations#

Commercial skincare products typically contain Matrixyl at higher concentrations than the 3 ppm used in the original clinical study, commonly in the range of 50-500 ppm. The raw material is supplied by Sederma as a solution that formulators incorporate into their products.

Application Guidelines#

For topical anti-aging use:

1. Cleanse the skin thoroughly
2. Apply the Matrixyl-containing product to the face and neck
3. Allow absorption before applying additional products
4. Use twice daily for optimal results
5. Combine with sunscreen during daytime application
6. Consistent use over at least 4-12 weeks is needed to observe visible results

Compatibility#
