Djillani et al. (2017) administered PE-22-28 at 3-4 mcg/kg intraperitoneally as a single dose in mice. This produced significant reduction of immobility time in the forced swimming test, with effects lasting approximately 23 hours (compared to 7 hours for the parent compound spadin).

Sub-Chronic Neurogenesis Protocol#

A 4-day sub-chronic treatment at 3-4 mcg/kg/day IP was sufficient to induce hippocampal neurogenesis and enhance synaptogenesis, as measured by increased PSD-95 expression in mouse cortical neurons. This rapid neurogenic effect compares favorably to traditional antidepressants, which typically require 2-4 weeks to induce neurogenesis.
