Research-Based Dosing Information#
Animal Study Protocols#
Obesity Study (2018)
The foundational study by Neelakantan et al. (PMID: 29155147) used the following protocol in mice:
- Species: C57BL/6 mice with diet-induced obesity (16 weeks high-fat diet)
- Route: Subcutaneous injection
- Dose: 20 mg/kg per injection
- Frequency: 3 injections per day
- Total daily dose: Approximately 34 mg/kg/day
- Duration: 11 days
- Group size: n=9 per cohort
Microbiome Study (2022)
Neelakantan et al. (PMID: 35013352) used 5-Amino-1MQ in combination with dietary modification in diet-induced obese mice. The NNMTi treatment was combined with switch from high-fat to low-fat diet.
Muscle Function Study (2024)
Dimet-Wiley et al. (PMID: 38969654) evaluated NNMTi in aged mice across four conditions: sedentary control, NNMTi alone, exercise alone, and NNMTi + exercise. Specific dosing details are in the published methods.
Critical Caveats About Animal Doses#
- Mouse doses cannot be directly converted to human doses using simple weight scaling. Allometric scaling, differences in metabolic rate, body surface area, and species-specific pharmacokinetics all affect translation
- The animal studies used subcutaneous injection, while many commercially available products are oral capsules. Oral bioavailability of 5-Amino-1MQ has not been characterized in published literature
- A published study on LC-MS/MS analytical methods for 5-Amino-1MQ in rat plasma (PMID: 34304009) exists, but does not establish oral bioavailability
- The HED (Human Equivalent Dose) calculator on this site can provide allometric dose conversion estimates, but these are mathematical approximations only, not clinically validated doses
Route of Administration#
In published preclinical studies, 5-Amino-1MQ has been administered by subcutaneous injection only. Despite this, the compound is commercially available in oral capsule form. This disconnect between the research evidence (injectable) and the market availability (oral) represents a significant gap -- oral bioavailability may be substantially different from injectable bioavailability, and no published study has characterized this.
